  • Bevacizumab Study in Early-Stage Colon Cancer Misses Primary Endpoint - OncologySTAT: Genentech, Inc., announced today that the National Surgical Adjuvant Breast and Bowel Project (NSABP) C-08 study did not meet its primary endpoint of improved disease-free survival (DFS) with adjuvant use of bevacizumab (AvastinŽ) plus chemotherapy in patients with early-stage colon cancer. This was based on results from the final analysis of the NSABP C-08 study, a randomized multinational phase III study designed to evaluate the efficacy of adjuvant 5-fluorouracil , leucovorin, and oxaliplatin (FOLFOX), with or without bevacizumab, in patients with resected stage II or III adenocarcinoma of the colon. Bevacizumab is already approved by the US Food and Drug Administration for the treatment of metastatic colorectal cancer. This is the first phase III study of bevacizumab in early-stage disease.

  • Competitor - Cisplatin - Wikipedia, the free encyclopedia - Paclitaxel adjuvant Taxol: Cisplatin is a platinum-based chemotherapy drug used to treat various types of cancers. It was the first member of its class, which now also includes carboplatin and oxaliplatin. Platinum complexes are formed in cells, which bind and cause crosslinking of DNA; ultimately triggering apoptosis(prog rammed cell death). Cisplatin combination chemotherapy is the cornerstone of treatment of many cancers. Initial platinum responsiveness is high but the majority of cancer patients will eventually relapse with cisplatin-resi stant disease. Many mechanisms of cisplatin resistance have been proposed including changes in cellular uptake and efflux of the drug, increased detoxification of the drug, inhibition of apoptosis and increased DNA repair. Oxaliplatin is active in highly cisplatin-resi stant cancer cells in the laboratory only to date. The drug Paclitaxel may be useful in the treatment of cisplatin-resi stant cancer; the mechanism for this activity is unknown.
